Evaluation of indirect immunofluorescent assay (IFA) by comparison with microscopic agglutination test (MAT) for swine leptospirosis diagnosis

Abstract

Leptospirosis is an important zoonosis and major economic lost problems in pigs. The recently standard method for diagnosis of Leptospira is a microscopic agglutination test (MAT). This method is sophisticated and difficult to apply in field practices. The objective of this study is to compare between MAT and Indirect immunofluorescent assay (IFA) methods. The reference Leptospira interrogans serovar grippotyphosa was reacted with 132 swine sera by both MAT and IFA. The result showed that all 30 negative MAT sera were negative with IFA. One hundred and one of 102 positive MAT sera were positive with IFA by titer ranging 1:25-1:150. The percentage of sensitivity and specificity of IFA comparative with MAT were 99.01 and 100 respectively. Therefore IFA can be used as screening test for swine leptospirosis.

โรคเลปโตสไปโรซีสในสุกร เป็นโรคติดต่อระหว่างสัตว์และคนที่มีความสำคัญและทำให้เกิดความสูญเสียทางเศรษฐกิจ การวินิจฉัยโรคเลปโตสไปโรซีสมาตรฐานในปัจจุบันใช้วิธี Microscopic agglutination test (MAT) แต่วิธีการดังกล่าวทำได้ยุ่งยากไม่เหมาะจะใช้ในพื้นที่ปฏิบัติงาน การศึกษาครั้งนี้จึงมีวัตถุประสงค์เพื่อเปรียบเทียบผลการวินิจฉัยโรคเลปโตสไปโรซีสในสุกรด้วยวิธี Microscopic agglutination test (MAT) และวิธี Indirect immunofluorescent assay (IFA) โดยทำการเปรียบเทียบระหว่างวิธี MAT และวิธี IFA โดยใช้เชื้อมาตรฐาน Leptospira interrogans serovar grippotyphosa ทำปฏิกิริยากับซีรัมตัวอย่างทั้งสิ้น 132 ตัวอย่าง ในกลุ่มที่ให้ผลลบด้วยวิธี MAT จำนวน 30 ตัวอย่าง ให้ผลลบกับการตรวจด้วยวิธี IFA ทั้งหมด กลุ่มที่ให้ผลบวกกับวิธี MAT จำนวน 102 ตัวอย่าง ให้ผลบวกกับวิธี IFA 101 ตัวอย่าง โดยมี IFA Titer อยู่ระหว่าง 1:25-1:150 ค่าร้อยละความไวและความจำเพาะของวิธี IFA เมื่อเทียบกับวิธี MAT เท่ากับ 99.01 และ 100 ตามลำดับ ฉะนั้นวิธี IFA สามารถใช้สำหรับการวินิจฉัยคัดกรองโรคเลปโตสไปโรซีสในสุกรได้
