Abstract

SSR molecular markers SSR are tandem arranged repeats of short DNA motifs (1-6 bp in length) distributed throughout the genome that frequently exhibit variation in the number of repeats at the locus of each rice variety. The SSR markers, therefore, are suitable for detecting polymorphisms between closely related genotypes of Thai local rice varieties from the National Rice Germplasm Bank. In this research, DNA fingerprinting was conducted using 8 Thai local rice varieties. i.e., Niaw Sanpahtawng (NSPT_GB), Khao Gaw Diaw (KGD_GB), Niaw Dam (ND_GB), Khao tah Haeng 17 (KTH17_GB), Jao’ Haw (JH_GB), Pin Gaew 56 (PG56_GB), Jek Chuey Gahb Khiaw (JCGK_GB), Sang Yod Pattalung (SYPL_GB), and 5 standard varieties, i.e., Khao Dawk Mali 105 (KDML105_GB, KDML105_C), Chai Nat 1 (CNT1_C), RD6 (RD6_GB, RD6_C), RD15_C, and Niaw Ubon 2 (NUBN2_C) which were used as control. Thirty two SSR markers, i.e., RM1, RM165, RM220, RM3627, RM157A, RM231, RM232, RM261, RM3, RM190, RM204, RM217, RM225, GT11, Glu23-23, RM248, RM149, B03, RM201, RM257, RM228, RM239, RM258, RM20B, RM21, RM202, RM206, RM209, RM224, RM17, RM20A and RM235 were used for DNA fingerprinting. DNA banding patterns of Thai local rice amplified with the 32 SSR markers showed different sizes of DNA fragments with 2-11 alleles. Using cluster analysis method, these 13 Thai rice varieties could be classified into 5 groups at the coefficient of similarity of 0.17.
